When creating events and then sending to notify people, can you upload a mailing list file?

No,

You would select the user group to whom you want to notify or Users from the website.

Is there a quick way to add new users to the website from a mailing list? With the acymailing newsletter I can upload from a mailing list. Would that be a quick way of adding emails from a separate mailing list?

Not that Jevents provides.

As when people use a calendar on a website they normally would also be frequent users of the site. This way gives much more control and functionality.

Do the emails have names/username within the list? if so... you could use Joomlapolis's Community Builder with CBJuice which would allow you to add the users to the website.
